Culture
medium: MEM
(Eagle) with Earle«s BSS, sodium bicarbonate, 2 mM L-glutamine, 1,0 mM sodium
pyruvate, 0,1 mM non-essential amino acids + 10% FBS
Subculture
routine: Centrifugation
and then resuspension. A concentration of 2 x 105 cells/ml is
optimal.
Incubate
at 37¡C, 5% CO2

Frequency Distribution 50 Cells: 2n=46.
This is a hypertriploid human cell line. The modal chromosome number is 78 occurring at 36%, with polyploidy at 5.9%. The marker chromosomes del(2)(pl3), der(19)t(6;19)(pll;pl2/13), der(19)t(9;19)(pl3;qll), der(17)t(7;17)(qll;pl3), and 12 others are common to most cells. The first 2 markers listed abouve are generally single-copied and teh remaining 3 are paired (four other markers not listed abouve also paired). N12, N14 and N17 are mostly single-copied, wheras N15, N16, and N19 are 4-copied. The C and Y chromosomes have two copies each and the Y«s are further confirmed in QM-stained preparations.
