Cell
Description: human
glioblastoma; astrocytoma; brain
Culture Medium: MEM
Eagle with Earle's BSS, sodium bicarbonate, 2mM L-glutamine, 1.0mM sodium
pyruvate, 0.1 mM non-essential amino acids + 10% FBS
Subculturing: Remove
medium, rinse with fresh 0.025% trypsin solution, remove trypsin and let the
culture sit at room temperature (or at 37¡C) until the cells detach (about 10
minutes). Add fresh medium, aspirate and dispense into new flasks.
Morphology: epithelial
Growth Properties: monolayer
Split Ratio: A
ratio of 1:2 to 1:5 is recommended
Fluid Renewal: 2
to 3 times weekly
Sterility: Tests
for Mycoplasma, bacteria and fungi were negative
Biosafety Level: 1
Tumorigenic: yes,
in nude mice
Karyotype: Chromosome
Frequency Distribution 50 Cells: 2n = 46. The stemline chromosome number is
hypodiploid, the 2S component occurring at 5.4%. The following nine markers
were common to most S metaphases: t(1q;?3), t(1pl22q), t(6p;?11q-), (6q;?7p),
t(7q;?), del(12q), t(20;1p;9q), t(?8p;?), and M1. Neither homogeneously
staining regions (HSR) nor double minutes (DM) were detected; all S metaphases
were monosomic for the X chromosome.
Antigen Expression: Blood
Type A, Rh+
Isoenzymes: Me-2,
1; PGM3, 1; PGM1, 2; ES-D, 1; AK-1, 1; GLO-1, 1; G6PD, B; Phenotype Frequency
Product: 0.0017
Depositor: Ponten
Description: This
is one of a number of cell lines derived from malignant gliomas (see also ATCC
HTB-15, ATCC HTB-16 and ATCC HTB-17) by J. Ponten and associates from 1966 to
1969.
Comments: established
from a 44-year-old caucasian female, grade III
ATCC No.: HTB-14
