Technical Information for Cell Cultures
Quality control
Before cell lines are accessed to the collection they undergo several
quality control procedures. Special attention is given to detection
of mycoplasma in cell lines as well as to bacterial and fungi testing.
These and other quality control are explained below.
Bacterial and fungi testing
Cell cultures and reagents are screened for the presence of bacteria
and fungi using casein-soya peptid, thioglycolat and sabouraud broths.
Mycoplasma testing and eradication
All cultures sent to inclusion in the collection are initially cultured
in antibiotic free medium and screened for mycoplasma using bisbenzimid
(Sigma B 1155) or DAPI (Sigma D 8417) fluorochrome dye. This direct
observation method has the advantages that it provides rapid results
and that the so called non-cultivable mycoplasma are easily detected.
When mycoplasma is detected it may not be possible to return to an
uncontaminated source of the cell line. In such cases eradication
may be attempted. Several methods are available including antibiotics.
