Cell line: RT-4

Cell description: RT-4 are derived from a recurring papillary tumor of the urinary bladder of a 63 year old male caucasian (Blood Type O+).
Morphology: epithelial-like
Tumorigenic in cheek pouch of steroid treated hamsters.
Sterility: test for Mycoplasma, bacteria and fungi were negative
Karyotype: (P174) Hyperdiploid and hypertetraploid with abnormalities including dicentrics, breaks, translocations and minutes. Isoenzymes: Me-2, 1; PGM3, 1-2; ES D, 1-2; AK 1, 1; GLO-1, 1-2 ; G6PD, B.
Phenotype Frequency Product: 0,0050
HLA cell line Phenotype: A25(10), 3; B12; Cw3;
Propagation: Culture Medium: Mc Coy's 5a medium, 90%; fetal bovine serum, 10%
Subculture Routine: remove medium, rinse with 0,25% trypsin solution, remove trypsin and let the culture sit at room temperature (or at 37°C) until the cells detach (about 10 minutes). Add fresh medium, aspirate and dispense into new flasks.
Subculture: every 6 to 8 days
Split Ratio: a ratio of 1:2 to 1:4 is recommended
Fluid Renewal: 2 to 3 times weekly
Growth Properties: monolayer
